It's about Roleplay in general, but also for rich settings like Dragonlance... what do you think...
How deep the lore knowledge of the DM before he dare to DM in such settings?

I'd say, depends on the DM.
For me, personally, setting is very important. Not just its lore and rules, but its themes. I'm generally not comfortable running a game in a setting I know little about. I'd have to do a lot of research beforehand. If there are novels or videogames for that setting, I like to experience them first so I have a better understanding of it. Failing that, at least a deep dive into whatever wiki it might have.
